During non-disjunction the homologous or sister chromatids do no separate due to malfunction in either Topoisomerase II, condensin, or separase. Due to absence of separation one of the cell lacks one chromosome and one other cell receives and extra chromosome making 2 cell abnormal having n-1 and n+1 karyotype respectively.
