We will do the staining of an endospore bacterium. We will do spore staining of Spore-forming bacteria include Bacillus (aerobic) or Clostridium (anaerobic) species. We will use Indian ink stain for the staining. Specially Saferin ( for gram negative ) and Crystal Violet ( for gram positive).
Because the gram-positive bacteria retain the crystal violet colour and stain purple whereas the gram-negative bacteria lose crystal violet and stain red. Thus, the two types of bacteria are distinguished by gram staining. Gram-negative bacteria are more resistant to antibodies because their cell wall is impenetrable.
The main purpose of endospores staining is to differentiate bacterial spore formers from non-spore formers. Spores are resistant to heat, dessicated chemicals and radiation. Within bacteria, endospores are protective structures used to survive extreme conditions but this protective nature makes them difficult to stain using normal techniques such as simple staining and Gram staining.
The India ink stain is used to stain capsules. Capsules appear as a well defined clear zone around the cell. Capsule formation may be evident in smears from clinical specimens, but not when the organisms are grown on most common laboratory culture media.
