Gram staining is usually the first differential stain used with bergey's manual to identify bacterial cells.

The structural differences between bacterial cells (prokaryotic cells) and eukaryotic cells are discussed. They are made up of numerous cytoplasmic inclusions, the bacterial chromosome, different cell surface features, the cell wall, and the plasma membrane (nucleoid). Except for a few, not all structures are seen in every genus.

A chemically intricate cell wall almost always surrounds bacterial cells. A periplasmic space divides the plasma membrane from the cell wall, which is located inside the former. Mesosomes are uncomplicated internal membrane structures that can be formed when this membrane is invaded.
