This is between L3 and L4 and L4 and L5 of the lumbar Vertebrae of the vertebral column. The medical procedure is  also called Lumbar Puncture, vertebral puncture.

The fluid collected is called Cerebrospinal fluid (CSF).It acts as shock absorber for the brain and spinal cord. It also contains some nutrients for these organs.

Procedure.

The patient or subject must lie  by his side, and the knees draws up towards the chest. Alternatively, the subject can sit up or bent forward so that the vertebral column is bent  and locations above  revealed.

Then anesthesia is performed at the lower spine to neutralize the pain of the spine needle.

Finally, the spine needle is inserted between the L3 and L4 or L4 and L5 to draw a clear liquid; mostly water. This is CSF

The procedure may also be used to determine the pressure in the brain.
